北宋仁宗年间，闻听开封府有人击鼓喊冤，兰桂坊女大厨胜男（姜鸿饰）策马飞奔去为民请命。柳明月（佘诗曼饰）乃居家过日子的妇道人家，家中有儿子和丈夫，虽与胜男出身不同、性格迥异，俩人最近都被莫名卷入连环命案。在开封府，一般断案是靠包大人（方舟波饰）和其手下得力助手南侠展昭（王翔弘饰），而北侠欧阳冬（欧阳震华饰）也是智勇双全的人物。自明月与胜男卷入命案之后，她们各展自身所长，帮助朝廷破案，仁宗帝下旨，封她们为“带刀女捕快”，成为包拯的左膀右臂，协助开封府衙。在展昭、公孙策（关菁饰）等人协作下，竟磕磕碰碰破了不少疑难案件。虽然在办案过程中笑话迭出，但正义与良知使她们担负起了除暴安良的使命。
